You can try, but you will only get 4 keys. Why you scale down where you stay (as you are not going to spend that much time in your rooms, heck, it’s Vegas) and get an additional room.
Also, unless you book through a hotel directly for the flight, I would book separately as the hotels are really hurting and rates fall all the time. Plus, if rates fall, you can cancel and rebook.

Here is the official policy for all Las Vegas hotels.
1) Rates are based on single or double occupancy. An additional fee for the 3rd and 4th party occupants will be charged, per night.
2) Maximum occupancy per room is 4 persons, regardless of age.

With that said, you can book a suite and have 5 adults there but if you get caught, they will bill you for the additional people.
People do what you are trying to do all the time. Here are ways you can get away with it.
1) Only two of you should approach the front desk at check-in
2) Don’t ask for additional room keys
3) Do not make noise in your room /no parties
4) Do not request extra towels
5) The security guards will let all of you in the elevator if one of you have a room key
6) Be ready to pay if you get caught

It would be best to get two rooms and ask for adjoining rooms. You will have two rooms with a door between the rooms like making it into one big room. That would be best and safest way to do it. Not work chancing stuffing everyone into one room and then get caught. Housekeeping will know how many people are staying in the room just by the condition of the room when she cleans it. Book two adjoining rooms.

You could possibly get away with 5 in the room. The maid won’t care. Tip her daily. But with 2 queen beds in the room, it would be difficult putting 3 in a bed. Also, if you book an air/hotel package you won’t get away with it. Only if you book hotel separate. Check out staying at the Rio, all rooms are suites and is reasonable. It has beds plus a living room w/ sofas.

Five in a room would really suck. There isn’t enough room. If you do not pay the extra for the additional people, you will be thrown out. The elevators, hallways and stairwells are video monitored and the housekeeping staff keep an eye out as well.
You would be far better off with two adjoining rooms. Paying an extra fee, for the extra person and getting a bed and bedding for that person. Adjoining rooms have a common door between them that can be locked from both sides. When you see how much more comfortable it is you will be glad you did.
